Preparation of Nutrient Broth
Step 1: Prepare the Nutrient Broth
In a clean container, mix 500 ml of distilled water with 10 g of glucose, 2 g of yeast extract, and any additional nutrients required for your specific fermentation culture. Stir until fully dissolved.
Step 2: Autoclave the Nutrient Broth
Transfer the nutrient broth solution into a sterile fermentation vessel. Autoclave the vessel at 121°C for 15 minutes to sterilize the broth.
Inoculation and Fermentation
Step 3: Cool and Inoculate
After autoclaving, allow the broth to cool to room temperature. Once cooled, add 5 g of the fermentation starter culture to the nutrient broth using a sterile pipette to prevent contamination.
Step 4: Monitor pH
Use a pH meter to check the pH of the broth. Adjust the pH to the optimal level for fermentation (typically around 6.5) by adding small amounts of acid or base as necessary.
Step 5: Incubate
Place the fermentation vessel in an incubator set to the optimal temperature for your fermentation culture (usually between 25-37°C). Allow the mixture to ferment for 24-48 hours, depending on the specific culture being used.
Harvesting
Step 6: Harvest the Culture
After the fermentation period, check for bubble formation or sediment, indicating fermentation activity. Use a sterile pipette to collect samples for further analysis or application.
Step 7: Store or Use
If not using immediately, store the culture in a sterile container at 4°C. If using, follow with the desired application in your research or production process.
